    ISOLATION AND IDENTIFICATION OF PASTEURELLA MULTOCIDA FROM LAYER AND PRODUCTION OF INFECTION IN EXPERIMENTAL BIRDS

    Abstract
    For the isolation and identification of Pasteurella multocida, a total number of 144 samples (liver, spleen and heart) collected from 4(four) Upazilas of Sirajgonj district. The samples were collected aseptically and transported to the laboratory of the department of microbiology, Faculty of Veterinary and Animal science, Hajee Mohammad Danesh Science and Technology University, Dinajpur.The samples were cultured on various culture media for the growth of the organisms. In nutrient broth, the organism produces turbidity within 24 hours of inoculation into incubation at 37° C. On nutrient agar media, it produces whitish, opaque, round, flat, translucent colonies of sticky mucoid or dry consistence of about 1-3 mm in diameter. On blood agar media it produces characteristics colonies of Pasteurella multocida .The organism produces whitish, opaque, circular and translucent colonies without hemolysis. On Eosine Methylene Blue agar, the organism produces small, circular, smooth, convex, translucent, glistening colonies which showed the tendency to coagulase. It did not grow on MacConkey agar. The organism was also characterized by gram’s stainining, leishman staining and methylene blue staining of the impression smear of liver and heart blood. Pasteurella multocida organism was also identified by biochemical test with 5 basic sugars where it fermented dextrose, lactose and mannitol wth the production of only acid but did not ferment maltose and lactose. In MIU, it was found non-motile, Indole positive and Urease negative. On TSI it produced H)S and fermented only glucose. It was MR negative and VP negative. Among 144 samples, 53 were found positive (36.80%).For the production of infection in experimental commercial layer birds, 1ml Pasteurella multocida suspension containing 62x10° CFU was inoculated orally.
