dc.description.abstractInvestigation was carried out in the Tissue Culture Laboratory of the Department of Genetics and Plant Breeding, Hajee Mohammad Danesh Science and Technology University, Dinajpur during November, 2011 to May 2012 with a view to study in vitro regeneration of Brassica spp. from explants, for this purpose, the three genotypes of Brassica viz. BARI Swisha-9. BARI Sarisha-11 and Tori-7 were used to assess their regeneration ability. Leaf segments of the three genotypes of Brassica were cultured on MS medium with different concentrations and combinations of hormones, range of percent callus induction was 55-100%. Among the three genotypes, BARI Sarisha-9 genotype showed early callusing (6.22 days) with maximum of callus induction (55%). Early and maximum rate of callusing appeared in MS +1.5 mg/L 2, 4-D + 0.3 mg/L NAA 2.0 mg/L AgNO; for all genotypes. Shoot regeneration ranged from 50% to 78.33% and BARI Sarisha-9 had the highest percentage of shoot regeneration (73.88%). Early and maximum rate of regeneration was found in MS + 3.5 mg/L BAP + 0.5 mg/L + 2.0 mg/L AgNO; for all the genotypes. The highest number of roots per shoot was counted in BARI Sarisha-9 (76.67%), in % MS + 0.5 mg/L NAA. Considering the overall performance, genotype BARI Sarisha-9 appeared the callus formation, shoot regeneration and root formation.en_US
