dc.description.abstractAn investigation was carried out with four potato genotypes in CRD Design with four replications at tissue culture laboratory of genetics and plant breeding department, Hajee Mohammad Danesh Science and Technology University, Dinajpur, Bangladesh during October 2016 to July, 2017. The objectives of the research work were to find out the response of potato genotypes in different culture media using different hormones and to characterize the experimental material through five important characters to assess the degree of diver gene and association among the experimental potato genotypes. The observations were recorded on randomly selected plants per genotype in each replication. Analysis of variance indicated highly significant differences among the genotypes, treatments and their interaction for all the traits under study. Comparing the mean values of data collected in two weeks, four weeks and six weeks, the genotypes Asterix exhibited superior performances for most of the traits except germination percentage. Similarly the culture media using T1 treatment showed best results for most of the traits. The interaction effect of variety and treatment revealed that the variety Asterix performed statistically best in culture media using T1 treatments for root length; shoot length and number of node whereas Diamant showed best results for number of leaf. Germination percentages responded differently in different weeks of data recording. Character association studies revealed that the traits length of the dominant shoot and number of its nodes, number of leaf and number of nodes, root length and shoot length entirely showed significant positive association with each other in different weeks of data recording. Germination percentages showed non-significant correlation with most of the traits. This indicated that simultaneous selection of variety by considering the character association with all these traits is important. The present investigation revealed that Asterix variety showed better performance under in vitro condition in the culture media with T1 treatments.en_US
