BIOCHEMICAL AND HISTO-MORPHOLOGICAL CHANGES OF PANCREAS OF RABBIT AS EFFECT OF INSECTICIDE (DIAZINON) CONTAMINATED FOOD

Abstract
Diazinon is a common organophosphorus insecticide that is widely used in our country in the
form of Rizinon. The irrational use of diazinon in our country can play a crucial role in the
occurrence of many diseases of animals and human. The present study was conducted to
investigate the alteration of gross & histomorphology of various organs, biochemical
parameters of rabbit as effect of insecticide (Diazinon) contaminated food. For this purpose, a
total number of 16 male rabbits (4 months of old) weighting 1200-1400gm were assigned into
four dietary treatments. Control group received only basal diet without any supplementation.
The other groups T1, T2 and T3 received diazinon with supplemented feed at a dose level of
15, 30 and 45mg/kg feed respectively. The body weight of each rabbit was weighed at 3 days
interval and found significant variations among the groups. Remarkable decrease in weight
gain in T3 group was due to malabsorption of nutrients and disturbances of metabolism due to
toxic effect of diazinon on the liver. Blood and tissue samples were collected on the 43rd days
of dietary treatment. Diazinon caused elevation of SGPT/ALT (P<0.01), serum creatinine
level attributed to pathological lesions including enlarged, pale & friable liver, swollen
kidneys and congestion on intestinal tract in experimental groups. But there was no
significant increase of RBS (random blood suger). Histopathological lesions in liver included
cirrhosis and necrosis in only (T2 & T3) treated groups. In kidney, glomerulus was filled with
reactive cells in group T1.While fibrosis, necrosis and connective tissue proliferation in the
tubuler and peritubular structure and fatty degeneration & cytoplasmic vacuoles in the tubular
cells of kidney were found in groups fed diazinon at higher doses (T2 & T3).In pancreas, there
was slight loss of normal histological structure of pancrese in group T1, decreased beta cell
population in the islets of langerhans, necrosis & fibrous tissue proliferation in interlobular
septa and intralobular parenchyma of pancrease in group (T2 & T3) .It was concluded that
diazinon toxicity in rabbit feed had a dose dependent effect on body weight gain, biochemical
parameters, gross and histological changes.
